Rye berry preperation methods outlined in his five steps and use synthetic filter discs am I running the risk for contamination if I inoculate using a glove box? What I'm really asking I guess is do I have to unscrew the top and inoculate with a syringe or can can I mark where the holes are on the filter disc and stick the needle through the holes already made for gas exchange? Is that a no no?

With the jar lids you create one 1/2inch hole for a breather port and a nail sized hole for a inoculation port.

Use a bead of silicone over the inoculation hole and let it cure, when you put a syringe through it and pull it out the silicone seals itself back up thus (Self healing injection port)

You do not want to inoculate through the filter port itself.If I follow RR's - Mushroom Cultivation (31)

yes, you want to unscrew the top and squirt some spore solution against the glass.
you should not stick the needle through the synthetic filter discs, you will probably destroy the needle and most certainly destroy the SFD.

First you need to study a LOT more studying.

2nd, you need 1, 1/4'' hole in your lid for gas exchange. Take a little bit of polyfil and stuff the hole but not to tightly. You don't wanna break or bend your needle. Then place you filter disk over the lid and screw on rig.

EDIT: I didn't read this post correctly. I thought you were saying lift the lid and and squrit spores in. I apologize. Best of luck!

When using the whole SFD >>>>

Place the lid first >> SFD >> then the ring.

Sterilize

When knocking (Inside your SAB) Remove ONLY the ring, not the entire lid(rubber side down is best for this method)

lift the SFD slightly and knock through the holes.

Replace SFD >> jar ring

This way the jar is never actually opened.

You want to use these w/ GE hole and RTV silicone injection port:

that way you could use one SFD on 7 or so jars instead of one like RR does.

I've colonized hundreds of jars using 1/4", 3/8", and 1/2" GE holes... if it's a quart/pint, 3/8" will work best with cube myc on grains IMO/IME

Read through some of these links, they should answer all of your questions and then some...
